摘要
目的观察茯苓多糖硫酸酯(SP)及其L-鸟氨酸盐(SP-Orn)对腺嘌呤致大鼠肾衰竭的防治作用。方法通过ig腺嘌呤250 mg.kg-1.d-1,连续21 d,使大鼠产生类似于人的肾衰竭症状。造模第5天开始,动物分组,SP组ig SP 450 mg.kg-1.d-1,SP-Orn组ig SP-Orn 700 mg.kg-1.d-1,SP组与SP-Orn组为等摩尔量,同时设模型组和地塞米松阳性对照组,连续给药30 d。给药结束后,代谢笼动物收集12 h尿液,测定大鼠尿量、尿肌酐(Ucr)、尿蛋白含量;眼眶后静脉丛采血,测定血清肌酐(Scr)、血尿素氮(BUN)、血清总蛋白(TP)和白蛋白(ALB)含量;取肾脏作病理学检查。结果与模型组比较,SP和SP-Orn组尿量、Ucr排出显著增加,尿蛋白排出则明显减少;Scr和BUN水平有明显的降低,TP和ALB明显增加;血蛋白和尿蛋白水平已接近正常对照组。肾脏系数明显下降,肾小球、肾小管数目增加,远曲小管扩张程度明显减轻。总体上SP-Orn的防治效果好于SP。结论SP和SP-Orn对大鼠肾衰竭有明显的治疗作用,SP和L-Orn在保护肾衰大鼠肾功能上存在正向协同作用。
Abstract
OBJECTIVE To investigate the preventive and therapeutic effects of sulfated pachyman(SP) and its salt of L-ornithine(SP-Orn) on renal failure induced by adenine in rats.METHODS The rats were administrated with adenine to induce renal failure at a dose of 250 mg·kg-1·d-1 for 21 d.From the fifth day on,SP and SP-Orn were given to two groups treated with the doses of 450 and 700 mg·kg-1·d-1 respectively.After 30 d treatment,the urine excreted within 12 h was collected.The volume of urine,the contents of urinary creatinine(Ucr) and urinary protein were determined.Blood was drawn from the ophthalmic venous plexus and the contents of serum creatinine(Scr),blood urea nitrogen(BUN),total protein(TP) and albumin(ALB) were determined.The morphological changes of kidneys were observed.RESULTS Compared with those of model group,after 30 d treatment,SP and SP-Orn significantly increased the volume of urine,the contents of Ucr,as well as serum TP and ALB,and declined the contents of urinary protein,Scr and BUN.The levels of serum TP and urinary protein were nearly equivalent to those of normal control group.And also,SP and SP-Orn alleviated the injuries of the kidneys.As a whole,the therapeutic effect of SP-Orn was better than that of SP.CONCLUSION SP and SP-Orn appeared to have a promising therapeutic effects on renal failure.There was a synergetic effect between SP and SP-Orn on curing renal failure in rats.
