OBJECTIVE To study the effects of velvet antler polypeptides(VAP) on the proliferation of hepatic cells and liver regeneration.METHODS L-02 cell line and rat spleen cells were cultured in different concentration of VAP for 24 h.MTT colorimetry was used for assaying proliferation.Mice modles of liver partial hepatectomy was established to study the effects of VAP on liver regeneration.70 Mice were randomly divided into 5 groups.Mice in the VAP groups were treated with VAP at a dose of 12.5,25 and 50 mg·kg-1·d-1 for 3 d subcutaneously.At the same time,mice in the control group were given equivalent amount of 0.9% sodium chloride solution and mice in the positive control group were given 200 mg·kg-1·d-1 BDD subcutaneously.After 12 h of the last administration,all mice were sacrificed.The relative liver weight was obtained.RESULTS VAP 10 and 20 mg·L-1 VAP showed significantly proliferation-promoting activity for L-02 cell line and rat spleen cells and increased incorporation of MTT colorimetry from(0.969 8±0.071 6;0.810 8±0.075 1) Bq(control group) to(1.218 8±0.153 3;1.021 7±0.111 2) and(1.567 7±0.084 9;1.349 8±0.067 9) Bq,respectively(P<0.01 or P<0.001).Compared with the control group,VAP 50 and 25 mg·kg-1 significantly stimulated liver regeneration in mice excised partial liver(P<0.01).CONCLUSION VAP showed a remarkable enhance effect on the liver regeneration after hepatectomy by stimulating proliferation of hepatic cells.
