摘要
目的建立同时测定丹参药材及其制剂中丹参素、原儿茶醛、丹酚酸A、丹酚酸B4种酚酸类成分含量的RP-HPLC色谱法。方法色谱柱:InertsilC8-3柱;流动相:乙腈-水-甲酸(24∶76∶0.4);流速:1.0mL·min-1;检测波长:285nm。结果在此色谱条件下4种酚酸类成分可完全分离。丹参素、原儿茶醛、丹酚酸A、丹酚酸B的线性范围分别为:8~400(r=0.9998),4~200(r=0.9994),4~200(r=1.0000)和4~200mg·L-1(r=1.0000)。4种成分在药材和制剂中的回收率均在98.5%101.0%,RSD(2.0%。结论该方法简便、准确、快速,可同时测定丹参药材及其制剂中4种酚酸类成分的含量。
Abstract
OBJECTIVE To develop a HPLC method for the simultaneous determination of danshensu, protocatechuic aldehyde, salvianolic acid A, salvianolic acid B in Salvia miltiorrhiza Bge. and its preparations.METHODS The analysis was carried on a column of Inertsil C8-3 with a mobile phase of acetonitrile-water-formic acid(24∶76∶0.4) at the flow rate of 1.0 mL·min-1.The detection wavelength was 285 nm. RESULTS The linearity was obtained over the range of 8~400 mg·L-1(r=0.999 8) for danshensu, 4~200 mg·L-1(r=0.999 4) for protocatechuic aldehyde, 4~200 mg·L-1(r=1.000 0) for salvianolic acid A and 4~200 mg·L-1(r=1.000 0) for salvianolic acid B, respectively. The average recoveries of four phenolic acids in Salvia miltiorrhiza Bge. And its preparations determined were all in 98.5%~101.0%, and the RSDs were less than 2.0%. CONCLUSION The method is simple, accurate and rapid. It may be suitable for the usual quality control method of Salvia miltiorrhiza Bge. and its preparations.
