摘要

目的对白术(Atractylodes macrocephala Koidz.)的抗炎活性成分进行研究。方法采用正相、反相硅胶柱色谱对白术中的化合物进行分离,依据理化性质、波谱数据分析进行结构鉴定,采用整体动物试验,对各成分进行初步抗炎作用研究,结果从白术中分离鉴定5个化合物,分别为白术内酯Ⅲ(Ⅰ)、白术内酯Ⅰ(Ⅱ)、12-异戊烯酰-14-乙酰-2E,8E,10E-三烯-4,6-二炔-1-醇(Ⅲ)、12-α-甲基丁酰-14-乙酰-2E,8E,10E-三烯-4,6-二炔-1-醇(Ⅳ-1)、12-β-甲基丁酰-14-乙酰-2E,8E,10E-三烯-4,6-二炔-1-醇(Ⅳ-2)。抗炎结果表明,上述各成分对小白鼠急性炎症模型均有一定的抗炎作用。结论白术中含对小白鼠急性炎症模型有一定作用的抗炎活性成分,其中化合物Ⅲ及化合物Ⅳ-1 13C-NMR波谱数据为首次报道。化合物Ⅳ-2数据为首次报道。

Abstract

OBJECTIVE To study the constituents of Atractylodes macrocephala Koidz. and its anti-inflammatory activity.METHODS The chemical components were isolated by silica gel column chromatography. Their structures were identified by spectral analysis. The anti-inflammatory pharmacological experiments of the constituents were carried out in vivo.RESULTS Five compounds were isolated and their structures were established as atractylenolide Ⅲ(Ⅰ), atractylenolideⅠ(Ⅱ), 14-Acetoxy-12-senecioyloxytetradeca-2E,8E,10E-trien-4,6-diyn-1-ol(Ⅲ), 14-Acet- oxy-12-α-methylbutyl-2E,8E,10E- trien-4,6- diyn-1-ol (Ⅳ-1) and 14-Acet- oxy-12-β-methylbutyl-2E,8E,10E-trien-4,6-diyn-1-ol(Ⅳ-2). CONCLUSION There are anti-inflammatory constituents in Atractylodes macrocephalae. The 13C-NMR data of compound Ⅲ and Ⅳ-1 are reported for the first time.The data of Ⅳ-2 is reported for the first time.
