摘要

目的非水毛细管电泳法测定不同产地金果榄药材中巴马汀和药根碱含量。方法在石英毛细管(50.0 cm×75μm)中,以75 mmol·L-1醋酸铵-0.1%冰醋酸甲醇溶液为电泳介质,运行电压15 kV,柱温25℃,检测波长265 nm。结果巴马汀和药根碱分别在1.99~99.5 mg·L-1(r2=0.998 5)和2.26~113 mg·L-1(r2=0.998 5)的范围内线性关系良好;其平均加样回收率分别为101.9%(RSD=4.2%)和100.9%(RSD=3.5%)。结论本实验方法快速简便,准确可靠,适用于金果榄等药材中巴马汀和药根碱的含量测定。

Abstract

OBJECTIVE To establish a non-aqueous capillary electrophoresis method for the separation and determination of palmatine and jatrorrhizine in Tinospora capillipes Gagn.from different original area.METHODS An uncoated fused-silica capillary(50.0 cm×75 μm) was used for separation in this study.The mobile phase consisted of 75 mmol·L-1 ammonium acetate-0.1% acetic acid in methanol.The applied voltage was 15 kV,and the column temperature was maintained at 25 ℃.The chromatogram was detected at 265 nm.RESULTS The linearities were obtained over the range of 1.99~99.5 mg·L-1 for palmatine,and the range of 2.26~113 mg·L-1 for jutrorrhizine.The average recoveries were 101.9% for palmatine,and 100.9% for jatrorrhizine,respectively.CONCLUSION This method is simple,rapid and reproducible,and it is successfully employed for the determination of palmatine and jatrorrhizine in T.capillipes Gagn.and other Chinese herbs containing these alkaloids.
