OBJECTIVE To establish a RP-HPLC method for the determination of 1-deoxynojirimycin(DNJ)in Ramulus Mori.METHODS The separation was performed with a DIAMONSIC C18 column(4.60 mm×250 mm,5 μm)at room temperature,and the mobile phase consisted of acetonitrile-0.1% aqueous acetic acid(1∶1),at the rate of 1.0 mL·min-1.The DNJ marked with 9-fluorenylmethyl chloroformate(FMOC-Cl)was detected at excitation wavelength of 254 nm,emission wavelength of 322 nm by Waters 474 fluorescence detector.Three factor,i.e.reaction temperature,buffer pH and reaction time on derivation of DNJ,were studied by orthogonal design.RESULTS DNJ had a good linear range of 3.6~36 mg·L-1(r=0.999 9,n=5).The average recovery was 99.2% with RSD of 3.5%(n=5).The detection limit was 0.08 mg·L-1(S/N=3).The optimum condition of DNJ derivation included reaction for 50 min at 30 ℃ with pH 7 potassium borate buffer.There was rich DNJ in Ramulus mori,and the DNJ contents were significantly different according to the mulberry varieties,stage and ages.CONCLUSION This method is simple,rapid,accurate,sensitive and applicable for the determination of DNJ in Ramulus mori and it products.The DNJ can be accumulated in Ramulus mori.
