摘要

目的应用α1-酸性糖蛋白(α1-AGP)柱,建立9-羟基利培酮手性异构体的HPLC拆分方法,为研究9-羟基利培酮手性异构体药动学提供基础。方法在α1-酸性糖蛋白柱上,考察了流动相pH值、有机改性剂种类及比例、缓冲盐种类及浓度、柱温对9-羟基利培酮手性异构体分离的容量因子、分离度、分离因子的影响,以摸索最优的拆分方法。结果以50mmol·L-1醋酸铵(pH6.8)-异丙醇(96.5∶3.5)为流动相,流速0.9mL·min-1,柱温25℃,在α1-AGP柱上成功拆分9-羟基利培酮手性异构体。结论9-羟基利培酮对映体在AGP固定相上得到了完全分离且分离良好。

Abstract

OBJECTIVE To develop a HPLC method for the enantiomeric separation of 9-hydroxyrisperidone on an α1-Acid glycoprotein(α1-AGP)column for pharmacokinetics study of 9-hydroxyrisperidone. METHODS To obtain optimistic method for the enantiomeric separation of 9-hydroxyrisperidone, influences of the mobile phase pH value, the type and ratio of organic modifiers, and the type and concentration of buffer, column temperature on the chiral separation of 9-hydroxyrisperidone were investigated.RESULTS 9-hydroxyrisperidone enantiomers were separated on an α1-AGP chiral stationary phase column with a mobile phase consisting of 50 mmol·L-1 NH4 Ac(pH 6.8 )-2-propanol(96.5∶3.5 )at a flow rate of 0.9 mL·min-1. CONCLUSION 9-hydroxyrisperidone enantiomers are separated well on a α1-AGP column.
