摘要

目的观察瑞香狼毒提取物对人增生性瘢痕成纤维细胞的抑制作用,为烧伤后增生性瘢痕的治疗提供实验依据。方法体外培养烧伤后的人增生性瘢痕成纤维细胞,加入不同浓度的瑞香狼毒提取物,24h后观察细胞形态学,以乳酸脱氢酶(LDH)为指标观察细胞毒性,用MTT法检测其增殖活性。结果不同浓度的瑞香狼毒提取物均能改变成纤维细胞形态,抑制细胞增殖,浓度在500μg·mL-1以下无明显的细胞毒作用。结论瑞香狼毒提取物对人增生性瘢痕成纤维细胞具有抑制作用,并呈剂量依赖关系,具有防治增生性瘢痕的应用前景。

Abstract

OBJECTIVE To investigate the effects of Stellera chamaejasme L. on postburn hypertrophic scar-derived fibrob-lasts.METHODS Hypertrophic scar-derived fibroblasts were harvested and cultured in vitro.The proliferation activity of the fibroblasts were examined by MTT method. The cellular morphology, proliferation activity and the cytotoxicity of the drug were observed after 24 h of culture. RESULTS The fibroblast morphology was altered by any concentration of Stellera chamaejasme L. solution.Furthermore,the cell counts and the cellular proliferation activity were decreased obviously by the treatment of Stellera chamaejasme L. CONCLUSION The cellular morphology and proliferation activity of postburn hypertrophic scar-derived fibroblasts are inhibited by Stellera chamaejasme L. in dose-dependent pattern, and it will be a potential agent for the prevention and treatment of hypertrophic scars.
