摘要

目的研究反相高效液相色谱法测定藏药裸茎金腰中β-peltoboykinolic acid含量的方法。方法蒸发光散射检测器:载气流速1.6 mL·min-1;漂移管温度68℃。Hypersil ODS-2色谱柱;流动相:甲醇-水(90:10);流速:1.0 mL·min-1;柱温25℃。结果测定β-peltoboykinolic acid的回归方程为A=8.06×106c-1.92×105,r=0.997 9;线性范围为0.392~1.176μg·mL-1;平均回收率为97.24%;RSD=4.27%;检测限为78.40 ng(2倍的信噪比)。结论该方法简便、可靠、准确,具有一定的实用性,可以作为裸茎金腰中β-peltoboykinolic acid的含量测定方法。

Abstract

OBJECTIVE To develop a HPLC method for the determination of β-peltohoykinolic acid in Chrysosolenium nudicaule Bunge.METHODS The flow rate of carried-gas was 1.6 mL·min-1 and the drift-tube temperature was 68 ℃ .The mobile phase consisted of methanol-water(90:10) and column temperature was at 25 ℃ .RESULTS A good linearity was obtained in the range of 0.392~1.176μg(r=0.9979,A=8.06×106c-1.92×105) .The average recovery was 97.24% and the RSD was 4.27% .The detect limit (S/N = 2) was 78.40 ng for β-peltoboykinolic acid.CONCLUSION The method was simple,reliable,accurate and practical.
