摘要

目的建立一种同时检测人血清中卡铂、假尿苷含量的高效液相色谱方法。方法色谱条件:Spherigel-C18色谱柱;流动相:含0.5%乙腈的0.02 mol·L-1KH2PO4缓冲液;流速:0.95 mL·min-1;UV:230 nm。结果本法卡铂的线性范围为1~20mg·L-1,相关系数r=0.990 2,平均回收率96%,最低检测浓度0.5 mg·L-1;假尿苷的线性范围为1~16 mg·L-1,相关系数r= 0.998 6;平均回收率97%,最低检测浓度0.3 mg·L-1。结论本法具有样品处理简单、稳定性高等特点,经临床验证具有较好的实用性且易于推广应用。

Abstract

OBJECTIVE To establish a method for the determination of carboplatin and pseudouridine in human serum.METHODS The concentration of carboplatin and pseudouridine was assayed on a Spherigel-C18 column with a mobile phase consisting of acetonitrile-0.02 mol·L-1 KH2PO4(5:1 000)at a flow rate of 0.95 mL·min-1 and detected at 230 nm. RESULTS The calibration curve of carboplatin was linear over the range of 1~20 mg·L-1 (r= 0.990 2) .the average recovery of carboplatin was 97% . The limit of determination was 0.5 mg·L-1.The calibration curve of pseudouridine was linear over the range of 1~16 mg·L-1(r = 0.998 6). The average recovery of pseudouridine was 97% . The limit of determination was 0.3 mg·L-1. CONCLUSION This method is simple and highly stabile. Clinical application demonstrated that this method was practical.
