摘要
目的研究土槿皮乙酸(Pseudolaric acid B)诱导HeLa细胞凋亡的作用,探讨其抗癌作用机制。方法采用噻唑蓝(MTT 法)、形态学观察、DNA凝胶电泳及Western blot检测法。结果土槿皮乙酸可剂量-时间依赖性的诱导HeLa细胞凋亡,5μmol·L-1土槿皮乙酸处理24 h的细胞,Hoechst 33258荧光染色后有明显的凋亡小体出现;处理36 h的细胞,有明显的DNA ladder出现。Bcl-2蛋白表达量-时间依赖性地减少,Bcl-xL蛋白表达无明显变化,而Bax和p53蛋白表达量-时间依赖性地增加。结论土槿皮乙酸对HeLa细胞的杀伤作用主要通过诱导细胞凋亡,降低Bcl-2蛋白的表达和增加Bax,p53蛋白表达为其诱发HeLa 细胞凋亡的机制之一。
Abstract
OBJECTIVE To study the mechanism of pseudolaric acid B-induced apoptosis on HeLa cells. METHODS MTT, fluorescence microscope observation, DNA agarose gel electrophoresis and Western blot analysis were used. RESULTS Pseudolaric acid B induces HeLa cells apoptosis in a time-and dose-dependent manner. Apoptotic bodies were observed in pseudolaric acid B-treated HeLa cells. The expression of Bcl-2 was reduced time-dependency, and the expression of Bcl-xL protein had no change. On the contrary, the expressions of Bax and p53 were increased time-dependently. CONCLUSION The cytotoxic effect of pseudolaric acid B on HeLa cells was mainly due to apoptosis. Mitochondrial oncogenes and p53 might be involved in the apoptotic pathways of pseudolaric acid B-treated HeLa cells.
