摘要

目的建立天花粉蛋白的免疫检测方法,用于鉴别天花粉药材。方法用天花粉蛋白(trichosanthin,TCS)的对照品免疫家免,制备多抗,然后用免疫亲和柱纯化抗TCS抗体,建立竞争ELISA方法,检测天花粉真伪品中天花粉蛋白含量,同时以蛋白免疫印迹技术证实了竞争ELISA方法检测天花粉蛋白含量的特异性。结果不同产地的正品天花粉新鲜品、饮片和标本天花粉蛋白的含量与各伪品中天花粉蛋白含量差别非常显著。结论利用此方法可以鉴定天花粉的真伪。

Abstract

OBJECTIVE To establish an immunization detection method of Radix Trichosanthis.METHODS Control rabbits were immunized with TCS. Antiserum was prepared and then emulative ELISA method was developed. By means of immune affinity column,anti-TCS antibodies was identified for detecting the TCS concentrations of Radix Trichosanthis and its confusing varieties. Meanwhile, their extracts was electrophoresed using SDS-PAGE, and trichosanthin then was transferred to NC film for performing protein immuneblot assay to testify the specialties of antibodies. RESULTS The contents of trichosanthin in fresh Radix Trichosanthis, processed Radix Trichosanthis. and sample were significant different from the false. CONCLUSION Radix Trichosanthis can be identified using protein immurization derection technology.
