摘要
目的 研究植入用缓释氟尿嘧啶体外释放度与人体血药浓度的相关性 ,以建立其体外释放度检查的方法。方法 用紫外分光光度法检测,检测波长为265nm ,以盐酸溶液 (9→1000)为溶剂,用摇瓶法测定植入用缓释氟尿嘧啶在360h内的累积释放量;用HPLC测定血浆内的药物浓度,数理统计其释放度与人体内药动学参数的相关性。结果 植入用缓释氟尿嘧啶的体外释放度与人体内药动学参数相关性显著。结论 方法简便 ,准确 ,可作为植入用缓释氟尿嘧啶的质量控制方法。
Abstract
OBJECTIVE To study the relation between dissolution of fluorouracil sustained release implant and the concentration in human plasma.METHODS The detection wavelength was at 265 nm.The release of fluorouracil sustained-release implant in vitro was measured with the surging flask in 360h The fluorouracil concentration in human plasma was determined by a HPLC method.The relationship between the dissolution and pharmacokinetic parameters was analysed statistically.RESULTS A good relationship was obtained between the dissolution and pharmacokinetic parameters. CONCLUSION This method is simple, accurate and suitable for the quality control of fluorouracil sustained-release for implant.
