摘要

目的:观察丁基苯酞 (3-n-butylphthalide,NBP)对低糖低氧刺激后培养的胎鼠皮层神经细胞谷氨酸和5-HT释放的影响?方法:胎鼠皮层神经元以低糖低氧培养基培养造成低糖低氧模型;谷氨酸和5-HT以荧光分光光度法进行测定?结果:低糖低氧10h可使神经细胞谷氨酸的释放量增加,dl NBP(1和10μmol·L-1)和l-NBP(10μmol·L-1)可降低细胞培养基中谷氨酸的浓度,而同剂量的d-NBP对谷氨酸的释放无明显影响?dl-NBP和l-NBP亦可抑制由花生四烯酸 (100μmol·L-1)引起的神经细胞谷氨酸的释放?本实验结果还显示,dl-NBP(1和10μmol·L-1)及l-NBP(10μmol0·L-1)可降低低糖低氧后神经细胞培养基中5-HT的浓度?结论:dl-NBP和l-NBP可抑制谷氨酸和5-HT的释放,这可能是其抗脑缺血作用的机理之一?

Abstract

OBJECTIVE:To investigate the effects of dl-3-n-butylphthalide (dl-NBP),d-NBP and l-NBP on the release of glutamate and 5-hydroxytryptamine (5-HT) from cultured perinatal cortical neurons.METHODS:The cortical neurons were cultured with medium containing low glucose and bubbled with N2,Glutamate and 5-HT were determined fluorometrically.RESULTS:The results showed that dl-NBP (1 and 10μmol·L-1) and l-NBP (10μmol·L-1) reduced the glutamate release and the release of 5-HT from cultured cortical neurons induced by hypoglycemia/hypoxia for 10 h.d-NBP was shown no significant effect.dl-NBP and l-NBP also decreased the glutamater elease induced by arachidonic acid (100μmol·L-1).CONCLUSION:The inhibitory effects of NBP on the release of glutamate and 5-HT may be one of its action mechanisms in the treatment of cerebral ischemia.
