目的:研究血清中青藤碱的液-固净化技术和在家兔体内的动力学特征。方法:自制硅胶萃取柱及萃取仪,将血清中青藤碱净化后,点样于硅胶G-硅胶GF254 ( 2 : I )薄层板上,以氯仿-甲醇(19:2)为展开剂,在岛津CS-930上,用双波长(λs275 nm,λR 320 nm )锯齿法扫描测定。结果:血清中青藤碱的固相提取回收率为96.95%±s 7.59%,用薄层扫描法测绘了2只家兔静注青藤碱后的药物浓度-时间曲线。结论:自制萃取柱净化血清样品简便、有效;青藤碱的薄层扫描测定法灵敏、准确。
Abstract
OBJECTIVE: To study the purification of sinomenine in serum by the liquid-solid extraction and the pharmacokinetics of sinomenine in rabbits. METHODS: To sinomenine in serum was extracted by silica column which made in our laboratory and redissolved in CHC13 .The sample was applied on silica GF254 plate and developed with CHCI3- CH3OH(19:2).The spot was detected by CS-930 atλs=275 m,λR=320 nm. RESULTS: The average recovery was 96.95%±s7.59%for SPE(solid phase extraction) procedure in serum. The drug-time curve of sinomenine in rabbitswas determined. CONCLUSION: The SPE columns which made in our laboratory are simple and efficient for the purification of serum samples. The TLC is sensitive and precise for the determination of sinomenine.
