摘要
目的:建立血浆中葛根素的高效液相色谱测定方法,为含葛根素中药复方体内代谢研究提供基础。方法:血浆用0.58 mol· L-1高氯酸沉淀蛋白质,采用Shimpack CLC-ODS柱(5μ, 150 mm x 4.6 mm ),预柱为YWG-C18H37,流动相为甲醇-5 . 625 mol· L-1醋酸-水(25:3:72),流速为1.00 ml·min-1,检测波长为247 nm。结果:该法回收率91.0%,方法重现性RSD=0.93%(n=6)。结论:本文建立的高效液相色谱测定方法为动物体内葛根素的测定提供了一个可靠的方法。
Abstract
OBJECTIVE: To establish the determining method of puerarin in plasma with high performance liquid chromatography. METHOD: The 0.58 mol·L-1perchloric acid was used to precipitate the protein. Shimpack CLC-ODS column(5μ, 150 mm X 4.6 mm) was used as analytical column. YWG-C18H37 was used as pre-column. The mobile phase consisted of methanol-5 .625 mol · L-1acetic acid-water(25:3:72).The rate of mobile phase was 1 .00 ml. RESULTS: The recovery of this method was 91,0%,and the repeatability was RSD=0.93(n=6).CONCLUSION : The method could be used to determine puerarin in vivo.
