摘要

用高效液相色谱法测定维司力农,以甲醇-1mmol/L醋酸(50:50)为流动相,检测波长271nm。最低检出全为0.05ng(R/N,5:1),在供试溶液为1~200μg/ml范围内峰面积与浓度线性关系良好(r=0.9999),重现性好,RSD为0.74%,回收率为99.00%±0.29%(n=9)。本法可同时分离维司力在及其合成中间体,并用于考察维司力农的微量有关杂质。

Abstract

Vesnarinone,a new positive inotropic agent,was synthesized by Tominaga et al ofthe Otsuka Pharmaceutical Co. Ltd. Institute in 1982. Vesnarinone and its impurities by reversed-phase HPLC were determined chromatograapy consists of a Shim-pack CLC ODS column(150mmx6mmID)with methanol :1mmol/L acetic acid(50: 50)as the mobile phase at a flow rate of 0.8ml/min. Spectrophotomtric detection was at 271nm and column temperature at 25℃. The detec-tion limits of vesnarinone was 0. 05ng(R/N,5:1),and the calibration curve was linear(r=0.99998)in the concentration range of 1~200Ug/ml.RSD was 0.74%.The recovery was 99.00%±0.29%(n=9).Vesnarinone and its intermediate compounds could be separated well under this coudition.The assay of vesmarinone and the related impurities was developed.
