Abstract:OBJECTIVE To establish the fingerprint of Melandrium firmum Rohrb. by HPLC-UV. METHODS The analysis was performed on SunFireTM C18 (4.6 mm×250 mm, 5 μm) column. The mobile phase was acetonitrile (B) and 0.4% phosphoric acid (A) with gradient elution. The detection wavelength and column temperature were 253 nm and 40 ℃, respectively. The flow rate was 1.0 mL · min-1. The injection volume was 10 μL. The chemical attribution of the fingerprint was determined by reference substance comparison method. RESULTS The HPLC-UV fingerprints of 10 batches of M. firmum were calibrated with 21 common peaks. Among them, three flavone C-glycosides were identified. The similarities of the chromatograms of 10 batches of M. firmum were greater than 0.95. CONCLUSION The method is simple and reliable, which has satisfactory precision, repeatability and stability. It can be used for the quality control of M. firmum.
