Comparation and Analysis of Three Kinds of Components in Angelicae Sinensis Radix Pieces and Danggui Jianzhong Decoction by UHPLC-Q-TOF-MS

Abstract:OBJECTIVE To compare the composition changes of organic acid, benzophthalide and coumarin in Angelica Sinensis Radix pieces and Danggui jianzhong decoction by UHPLC-Q-TOF-MS. METHODS The column was Thermo Accucore C18 (2.1 mm×150 mm, 2.6 μm),the mobile phase was 0.1% acetic acid-water (phase A)-acetonitrile (phase B). The chemical components of Angelicae sinensis in Angelicae Sinensis Radix pieces and Danggui jianzhong decoction were identified by PeakView, and the ion peak intensity was recorded. The ion peak intensity was used as the basis for the comparison of component content. RESULTS A total of 44 compounds were identified,and one of the compounds could not be detected in Danggui jianzhong decoction. After the compatibility of Angelica sinensis, there was no difference in the content of 9 components,5 decreased and 29 increased. Among the organic acids,the content of components with strong acidity decreased and the content of other components increased. The contents of coumarins increased. There was no consistency in the content change trend of phthalide components. CONCLUSION The content of most components of Angelica sinensis increased after compatibility,which was related to the solubilization of colloidal solution or the solubilization of other components,except that some components also contributed to other prescriptions. The decrease of organic acids with strong acidity such as chlorogenic acid and ferulic acid may be related to the dissolution competition of organic acids.
