Structure Characterization and Immunological Activity of Two Rhodiola Polysaccharides

Abstract:OBJECTIVE To compare the structural characteristics and the difference in toll like receptor(TLR4) agonistic activity of Rhodiola crenulata and Rhodiola fastigiata polysaccharides. METHODSRhodiola crenulata polysaccharide (RCP) and Rhodiola fastigiata polysaccharide (RFP) were isolated and purified by water extraction and alcohol precipitation. UV, IR, HPGPC, GC-MS and other spectral methods were used to analyze the composition and structure of RCP and RFP polysaccharides; HEK-Blue hTLR4 cells were used to compare the TLR4 activation rate of RCP and RFP. RESULTS RCP is a homogeneous polysaccharide with a molecular weight of about 125 000, containing arabinose and galactose, with a molar ratio of 0.089:2.233; RFP is a heteropolysaccharide with a molecular weight of about 68 000/134 000, containing rhamnose, arabinose, and galactose, with a molar ratio of 0.646:1.148:2.206. At the concentration of 100 μg·mL-1, the TLR4 activation rate of RCP was 87.59%, and the TLR4 activation rate of RFP was 33.39%. CONCLUSION RCP components are more homogeneous than RFP, and TLR agonistic activity is stronger, which provides a reference for the research and development of Rhodiola polysaccharide components.
