Abstract
OBJECTIVE The pharmacokinetics of all-trans retinoic acid (RA) conventional and sterically stabilized liposomes in rabbits were studied and compared.METHODS By a randomized cross-over design, six rabbits were received a single dose of either RA conventional or RA sterically stabilized liposomes intravenously. The RA in plasma was detected by high-performance liquid chromatography and the pharmacokinetic parameters were calculated by 3P87 pharmacokinetic programme.RESULTS The t1/2 of RA conventional liposomes and the sterically stabilized liposomes were 6 and 16 min.t1/2 47 and 247 min,MRT 51 and 133 min,AUC 171.2 and 391.9 gminmL-1, respectively. There was statistic significant difference in parameters between the two dosage forms.CONCLUSION The results showed that RA sterically stabilized liposomes had longer circulation time in plasma with much higher concentration than RA conventional liposomes.
