Abstract��
OBJECTIVE High performance liquid chromatography with column switching was developed for the determination of theophylline in plasma of dog. METHODS The plasma samples were injected onto a pretreatment column packed with Lichromprep the RP 18(25��40 ��m)after simple dilution.Distilled water was used to wash out protein and other impurities in plasma.After switching,the concentrated theophylline was eluted in the back-flush mode onto a Shimpack CLC-ODS column with methanol-water (22��78)as mobile phase.Purge solutions were used for clean-up and for regenerating the pretreatment column. RESULTS The limit of detection was 0.001 mg��L-1plasma.The linear range was 1��32 mg��L-1.The ��RSD�� of intra-and inter-day were less than 3.37% and 5.36% for theophylline in dog plasma,respectively. CONCLUSION The method was successfully used to determine the pharmacokinetics of theophylline sustained release tablets in 12 dogs.
