Quality Evaluation of Forsythiae Fructus from Different Habitats Based on Simultaneous Determination of Multiple Bioactive Constituents Combined with Multivariate Statistical Analysis

Abstract:OBJECTIVE To comprehensively evaluate the quality of Forsythiae Fructus from different habitats based on simultaneous determination of multiple active components combined with multivariate statistical analysis. METHODS Twenty-one bioactive constituents including phenylethanoid glycosides, lignans, flavonoids and phenolic acids in Forsythiae Fructus from different producing areas were simultaneously determined by ultra-fast liquid chromatography coupled with triple quadrupole/linear ion trap tandem mass spectrometry (UFLC-QTRAP-MS/MS). Based on the contents of 21 target components, hierarchical cluster analysis (HCA), principal component analysis (PCA) and One-way analysis of variance (One-way ANOVA) were further employed for the quality assessment of Forsythiae Fructus from different habitats. RESULTS The HCA results showed that 14 batches of medicinal materials from three major producing areas were divided into two categories: the first category was Lingchuan in Shanxi, Heyang in Shaanxi, Huixian in Henan, Linchuan in Henan and Luoyang in Henan; the second category was Anze in Shanxi and Guxian in Shanxi. The PCA results revealed that the comprehensive quality of Forsythiae Fructus from Shanxi was better than those from Shaanxi and Henan. Additionally, the results of One-way ANOVA indicated that the contents of 10 components, ie chlorogenic acid, ferulic acid, astragalin, arctiin, luteolin, forsythiaside B, forsythiaside A, pinoresinol-4-O-β-D-glucoside, phillyrin and pinoresinol were significantly different among the above three provinces. CONCLUSION The integrated evaluation system based on simultaneous determination of multiple bioactive constituents combined with multivariate statistical analysis is successfully applied to the quality evaluation of Forsythiae Fructus from different habitats. It can provide a new reference method for the quality evaluation and control of Forsythiae Fructus, which can also provide basic information for further expounding the geo-authentic of Forsythiae Fructus.
