FOXO1在肝脏糖脂代谢中的作用

Effect of FOXO1 on glucose and lipid metabolism in liver

摘要: FOXO1作为叉头转录因子家族中的成员,受到蛋白质间相互作用和磷酸化、乙酰化和泛素化等翻译后修饰。FOXO1能够促进糖异生基因G6Pase和PEPCK的表达,同时可结合到ApoC-III启动子上,促进ApoC-III的表达,加重高三酰甘油血症。FOXO1的磷酸化有助于促进肝脏MTP表达和VLDL产生。FOXO1在肝脏的糖脂代谢中的作用提供了胰岛素抵抗、糖尿病治疗的新的靶点。

Abstract: As a member of the forkhead box transcription factor (FOXO) family, FOXO1 is subjected to protein–protein interactions and post-translational modifications such as phosphorylation, acetylation and ubiquitination. FOXO1 can promote the expression of glucose-6-phosphatase (G6Pase) and phosphoenolpyruvate carboxykinase (PEPCK), and can bind to the apolipoprotein C-III (ApoC-III) promoter to induce ApoC-III expression and aggravate hypertriglyceridemia. FOXO1 phosphorylation contributes to the promotion of liver microsomal triglyceride transfer protein (MTP) expression and very low density lipoprotein (VLDL) production. The role of FOXO1 in glucose and lipid metabolism in the liver provides a new therapeutic target for insulin resistance and diabetes.
