BEZ235体内外抑制人乳腺癌细胞系增殖

BEZ235 inhibits the proliferation of breast cancer cells in vitro and in vivo

摘要: 目的 探讨BEZ235体内外抑制乳腺癌细胞的作用。方法 选用人三阴乳腺癌细胞系MDA-MB-231及三阳乳腺癌细胞系MCF-7,用MTT检测细胞增殖;用裸鼠荷瘤实验分析乳腺癌细胞系体内增殖;用Western blot法检测PI3K/Akt相关信号通路蛋白的表达。结果 BEZ235对乳腺癌细胞具有明显的增殖抑制作用(p<0.01),尤其是对三阴性乳腺MDA-MB-231细胞具有较强的体内外抑制作用(p<0.01)。结论 BEZ235可体内外抑制乳腺癌细胞,其机制可能是通过抑制PI3K/Akt信号通路蛋白的磷酸化修饰而实现的。

Abstract: Objective To investigate the effect of BEZ235 on breast cancer cells. Methods Proliferation ability of human triple negative breast cancer cell line MDA-MB-231 and triple positive breast cancer cell line MCF-7 was detected by MTT assay; the xenograft model to analysis the ability of exhibited breast cancer proliferation in vivo; Western blot method to detect the PI3K/Akt signaling pathway related protein level. Results BEZ235 significantly inhibited the proliferation of breast cancer cells (p<0.01). Especially BEZ235 can strongly inhibited the triple negative breast cancer MDA-MB-231 cells in vivo and in vitro (p<0.01). Conclusion BEZ235 may through the inhibition of PI3K/Akt signaling pathway proteins phosphorylation to inhibition of breast cancer cells in vitro and in vivo.
