人肺癌组织中MCPH1基因过表达后提高化疗药物抑制肺癌A549细胞的增殖

摘要: 目的 检测人肺癌组织中MCPH1基因表达,并用MCPH1真核过表达载体,检测其协同化疗药物对人肺癌细胞A-549的影响。方法 用Real-time PCR法检测肺癌标本中MCPH1基因相对表达量。用前期已构建的真核表达质粒(pcDNA3.1(-) / MCPH1)及对照空白质粒(pcDNA3.1(-))转染A-549细胞,实验分为3组:实验组(OVER)、空白对照组(W/O)、未处理组(NC)。然后分别用Real-time PCR和Westen blot检测转染后细胞基因转录和蛋白表达。结果 肺癌组织中MCPH1基因表达低于正常组织(P<0.05)。在肺癌A549细胞中分别加入阿霉素1.0μmol/mL、紫杉醇20μg/mL、顺铂0.1μg/mL,48h后转染MCPH1组抑瘤率明显高于对照组及未处理组(P<0.05)。结论 肺癌组织中,MCPH1基因表达下调。MCPH1基因过表达后协同化疗药物抑瘤率明显升高。

Abstract: Objective To detect MCPH1 mRNA expression in lung cancer, and over expression of MCPH1 in A-549 cells with drugs. Methods MCPH1 mRNA expression in lung cancer was measured by Real-time PCR.Eukaryotic expression plasmid pcDNA3.1(-)/MCPH1 and Blank plasmid pcDNA3.1(-) were transfected into A549 cells,and set three groups according to the different transfected plasmid: Experimental group (OVER)、Blank control group (W/O)、Untreated (NC). The MCPH1’S mRNA and protein expression were detected by quantitative Real-Time PCR and Western bolt. Results MCPH1 gene expression in cancer tissues is lower than normal tissues (P<0.05). In A549 cells,experimental groups were added respectively 1.0μmol/mL ADM, 20μg/mL TAX, 0.1μg/mL DDP ,48h later, The inhibition rate of experimental groups was significantly higher than the control groups and the untreated groups (P<0.05). Conclusion MCPH1 genes downregulated in lung cancer;the tumor inhibitory rate increased after MCPH1 gene overexpression concurrent chemotherapy drugs.
