FK506对NIT-1细胞增殖和胰岛素分泌的影响

Effect of FK506 on NIT-1 cells proliferation and insulin secretion

摘要: 目的 探讨他克莫司(FK506)对NOD鼠胰岛β细胞株(NIT-1)增殖、凋亡和胰岛素分泌的影响。方法 用MTT法、流式细胞术检测NIT-1细胞增殖;放射免疫法、Real-time PCR检测NIT-1细胞胰岛素分泌和胰岛素分化相关基因PDX-1、GLUT2和GK mRNA的表达。结果 FK506(20μg/L)抑制NIT-1细胞增殖(P<0.05),诱导细胞凋亡(P<0.05);FK506(10 μg/L)抑制NIT-1细胞释放胰岛素(P<0.05),胰岛素分泌相关基因PDX-1(胰岛素促进因子-1)、GLUT2(葡萄糖转运载体2) mRNA表达下调(P<0.05)。结论 FK506(10μg/L)可抑制NIT-1的增殖,诱导凋亡,可通过下调PDX-1和GLUT2 mRNA表达,抑制NIT-1胰岛素分泌。

Abstract: Objective To investigate the effects of FK506 on cell proliferation of pancreatic NIT-1 beta-cell and insulin secretion function. Methods Effect of FK506 on proliferation and apoptotic were mensured by MTT-assay, Flowcytometry; Insulin secretion was investigated by Radioimmunoassay and Real-time PCR. Results FK506 (20μg?L-1) could inhibit the proliferation of NIT-1 cells, and could induce apoptosis; FK506(10μg?L-1) could inhibit NIT-1 cell insulin release, The mRNA expression of PDX-1 and GLUT2 were down-regulated. Conclusion FK506 inhibit NIT-1 cells insulin secretion, the mechanism maybe related to the down-regulation of. PDX-1 and GLUT2.
