一氧化氮对大鼠背根神经节瘦素长型受体表达的影响

Eeffect of nitric oxide on the expression of long type of leptin receptor in dorsal root ganglion

摘要: 目的 研究一氧化氮(NO)对大鼠背根神经节(DRG)神经元中瘦素长型受体(OB-Rb)mRNA和蛋白表达的影响。方法 用一氧化氮供体硝普钠(SNP;0、 0.01、0.1和0.5 mmol/L)处理体外培养大鼠DRG神经元;用RT-PCR和免疫印迹技术,检测神经元中瘦素长型受体mRNA和蛋白的表达。 结果 0.1~0.5 mmol/L 的SNP能促进瘦素长型受体mRNA和蛋白表达。 结论 NO对DRG的功能调节,可能与瘦素相关。

Abstract: Objective To investigate the effect of nitric oxide (NO) on the expression of long type of leptin receptor (OB-Rb) in dorsal root ganglion (DRG) neurons cultured in vitro. Methods Sodium nitroprusside (SNP; 0, 0.01, 0.1 and 0.5 mmol/L) were used to treat the DRG neurons cultured in vitro. RT-PCR and Western Blot were used to detect the expression of long types of leptin receptor (OB-Rb) in DRG neurons. Results 0.1~0.5 mmol/L SNP can increase the expression of OB-Rb mRNA and protein Conclusion The modulation of NO on DRG neurons may be relative with the action of leptin.
